Long-term cognitive deficits after traumatic brain injury associated with microglia activation

چکیده

Traumatic Brain Injury (TBI) is the most prevalent of all head injuries. Microglia play an essential role in homeostasis and diseases central nervous system. We hypothesize that microglia may a beneficial or detrimental TBI depending on their state activation duration. In this study, we evaluated whether results spatiotemporal change phenotype it affects sensory-motor learning memory functions male C57BL/6 mice. used panel neurological behavioral tests multi-color flow cytometry-based data analysis followed by unsupervised clustering to evaluate isolated from injured brain tissue. characterized several microglial phenotypes association with cognitive deficits. increase activated correlated negatively spatial at 35 days post-injury. After injury, frequency increases intensity. Activated found brains correlate positively deficit post injury.

P108: Microglia in Traumatic Brain Injury

Microglia is one of the first innate immune components. These cells account about 5 to 10% of the entire adult brain cells and are activated by trauma. Complex-mediated inflammatory responses occur through cellular and molecular events during and after the traumatic brain injury (TBI). In-lesion area astrocytes, microglia, and damaged neurons begin to secrete cytokines and chemokines. Microglia...

Closed-head minimal traumatic brain injury produces long-term cognitive deficits in mice.

Victims of minimal traumatic brain injury (mTBI) do not show clear morphological brain defects, but frequently suffer lasting cognitive deficits, emotional difficulties and behavioral disturbances. In the present study we adopted a non-invasive closed-head weight-drop mouse model to produce mTBI. We examined the effects of 20, 25, or 30 g weight drop 7, 30, 60 and 90 days following injury on mi...

Brain biomarkers and pre-injury cognition are associated with long-term cognitive outcome in children with traumatic brain injury

BACKGROUND Children with traumatic brain injury (TBI) are frequently at risk of long-term impairments of attention and executive functioning but these problems are difficult to predict. Although deficits have been reported to vary with injury severity, age at injury and sex, prognostication of outcome remains imperfect at a patient-specific level. The objective of this proof of principle study ...

Age at injury is associated with the long-term cognitive outcome of traumatic brain injuries

INTRODUCTION The association between age at injury (AAI) and long-term cognitive outcome of traumatic brain injuries (TBI) is debatable. METHODS Eligible participants with a history of TBI from Alzheimer's Disease Neuroimaging Initiative were divided into a childhood TBI (cTBI) group (the AAI ≤ 21 years old) and an adult TBI (aTBI) group (the AAI > 21 years old). RESULTS The cTBI group has ...

Hyperoxemia and long-term outcome after traumatic brain injury

INTRODUCTION The relationship between hyperoxemia and outcome in patients with traumatic brain injury (TBI) is controversial. We sought to investigate the independent relationship between hyperoxemia and long-term mortality in patients with moderate-to-severe traumatic brain injury. METHODS The Finnish Intensive Care Consortium database was screened for mechanically ventilated patients with a...
